First, be aware, that you&#039;ll be growing unknown microorganisms in fairly large quantities, so take your safetly seriously - wear gloves, a mask, and eye protection while working with your cultures, wash your hands thoroughly after your experiments, and disinfect everything with bleach after you&#039;re done.  
Biofilms grow best on rough, hydrophobic surfaces.  The easiest way to grow manageable sized biofilm samples would be to suspend identically sized pieces of plastic (cut up soda bottles, cut up ice cream buckets, or whatever you have lying around) in your fish tank culture chamber, strung on fishing line/etc. like beads on a string.  After your biofilm is grown, you cantake each piece of plastic out separately for your treatments.  
If you&#039;re growing a bacterial biofilm, fill the fish tank with a dilute nutrient solution of commercial bacterial growth medium, chicken stock, or the like (this will get very stinky if you let it grow more than a day or two).  If you&#039;re growing an algal biofilm, put dilute fertilizer (say, 1/10 to 1/20 the amount recommended on the package) in your fish tank, inoculate the tank with algae from an existing fish tank, and  keep the tank in very bright light.  Either way, an air pump with air stone and an aquarium heater to raise the temperature will make your biofilm grow quickly.  
For your experimental samples, try several different times in the ultrasonic bath - say, 1 minute, 10 minutes, and 30 minutes.  Run at least three replicates at each time point.  I&#039;d use biofilm samples treated with a 10% household bleach as a positive control and biofilm samples treated with bottled water or aged tap water (to get rid of any chlorine from the water treatment plant) as a negative control.  
You don&#039;t need replica plating for this experiment - you&#039;re not trying to grow exact copies of a single colony of bacteria.  Instead, use a sterile bacteriological swab to swab a constant area (say 1 cm x 1 cm) of each piece of plastic, then transfer the bacteria to the agar plate with the swab using sterile technique.  
If you&#039;re looking at algal biofilm, don&#039;t bother with the agar plates - the algae won&#039;t grow well.  Just use visual inspection to score the amount of algae left on the plastic pieces instead.  </description>
